Effects of Insulin and Insulin-like Growth Factor-ion Ovarian Granulosa Cell Function in Catile
نویسندگان
چکیده
The direct effect of insulin and insulin-like growth factor-Ion ovarian granulosa cell function in cattle was evaluated by using a serum-free culture system. Granulosa cells were obtained from small (1 to 5 mm) follicles collected from cattle and cultured for 4 days. Culture medium was collected for quantification of progesterone, and cell numbers were determined. Both insulin and IGF-I significantly increased granulosa ceIl numbers and progesterone production after 2 days of treatment. We conclude that insulin and IGF-I may have direct effects on ovarian function in cattle.

Insulin and insulin-like growth factors (IGFs) have direct effects on cultured ovarian cells. These effects include stimulation of granulosa cell mitogenesis, granulosa and luteal cell progesterone production, and thecal cell androgen production and appear similar among species. However, species differences exist with regard to insulin and IGF-I effects on granulosa cell estradiol production. I...

In cells from both sizes of were to determine the effect of insulin, insulin-like follicles, insulin (1 pg/mL), in the presence of FSH, growth factor I (IGF-I), testosterone, and FSH on increased estradiol production per cell. In contrast, proliferation, progesterone production, and(or) esIGF-I (100 ng/mL) inhibited estradiol producti...
متن کاملInsulin like growth factor 1 and regulation of ovarian function in mammals.
Various growth factors have been proposed to play endocrine and/or paracrine role in mammalian ovarian follicular development. The insulin like growth factor 1 (IGF-1) is one such factor. More and more reports now support the existence of an intra-ovarian IGF system including receptors and binding proteins. The role of IGF-1 in ovary is to amplify gonadotropin hormone action in terms of increas...
متن کاملProduction and functional characterization of human insulin-like growth factor 1
Insulin-like growth factor 1 (IGF-1) is a polypeptide hormone produced mainly by the liver in response to the endocrine growth hormone (GH) stimulus. This protein is involved in a wide range of cellular functions, including cellular differentiation, transformation, apoptosis suppression, migration and cell-cycle progression and other metabolic processes. In the current study, human heart cDNA w...

The insulin-like growth factor I receptor (IGF-IR) plays an essential role in the establishment and maintenance of transformed phenotype. Interference with the IGF-IR pathway by antisense causes reversal of the transformed phenotype in many rodent and human tumor cell lines. We stably transfected the PC-3 human prostate cancer cell line with an IGF-IR antisense RNA expression plasmid. The ...
